Default Has Regulos only been banished once before..

by the Guardians, and the Defiant have yet to prove they are able to do this as well or that there is another way other than how the Guardians did so, by the giving of their life?

Do you mean that time when Eth learned to make powerful magitech, which was used in war against Blood Storm?

It was one of the Mkhai tribe who first drew energy from sourcestone, inspired by the ways the planes warp and change Telara to see the vast magic within the stone of creation. Building upon this breakthrough, we crafted engines of war to turn upon the dragons.

How they roared when our engines burned their armies to lakes of sludge, turned their champions into crystal statues, even singed the scales from the dragonsí mighty shoulders. We pushed the Blood Storm back across the desert, sealed many of them in tombs inescapable, and aided in banishing awesome Regulos.
Magitech was proven to be effective against Blood Storm. Also don't forget that magitech kept Port Scion shielded from forces of Endless Court - till Guardian Ascended sabotaged the defenses, that is.

On the other hand, alternate timeline of Defiant starting zone shows what happens when Guardians decide that they do not need no magitech.

Or do you mean the Guardians' starting zone?

is another way other than how the Guardians did so, by the giving of their life?
I don't think that "giving their life" is an essential component of banishment. It's rather a consequence of being caught in the side effects of destroying Regulos's container (Aedraxis's body). They could drop a magibomb on Aedraxis from the safe range, destroying the container and staying alive themselves. But of course, Guardians rather die or let the world be destroyed than use magitech.

Guardians didn't banish Regulos at first.

    Magitech, long ago, banished Regulos.

The Vigil (perhaps along with mages and magitech) created the Ward to keep him out.

All Guardian Ascended did was destroy Aedraxis' Regulos-infused body, causing such a shockwave that it ended the Rift, and thus banished Regulos' SPIRIT (instead of both his body and spirit), as well as shattering his body across Telara.

So only magitech has so far fully banished Regulos.
Guardian Ascended managed to banish Regulos' spirit again, but not his dragon body form.

(This is why in the Terminus future, we see Regulos in a humanoid 'Shade'(?) form, with his 'dragon spirit' roaring behind him.)

Regulos has only set foot on Telara twice.

The first time was during the Blood Storm wars, which had an abundance of magi-tech and predated every faction's existence.

The second time was during the Shade, which ended with an anonymous pre-Guardian hero braining Regulos with a really big hammer and getting owned in the resultant explosion.

thanks for these replies. I found myself trying to imagine, after listening to a conversation about how to defeat Regulos (if he comes again) what each side might have felt when they first became Ascended. I had forgotten about the first time Regulos was banished completely, spirit and form, and also that the fractions were created moreso after Aedraxis form was shattered by the Hammer of the Vigil
